Before establishing as a parish, Padukone was part of Immaculate Conception Church, Gangolli. Parish Priest of Gangolli Fr P.V. Gonsalves (1926-1937) built a chapel at Padukone. The chapel was raised to the status of parish on May 31, 1936 by the decree of Bishop Victor Fernades of Mangalore Diocese.
